Genistein has shown potential in the treatment of complex diseases such as cancer, diabetes, cardiovascular diseases, and menopausal symptoms. However, its limited solubility and bioavailability hinder its clinical application. To address this issue, a research team led by Lu Yang and Du Guanhua prepared a genistein-piperazine cocrystal using cocrystal engineering. The cocrystal's structure, stability, solubility, and bioavailability were confirmed through various analytical techniques. The results demonstrated that the cocrystal improved solubility and bioavailability compared to previous studies. This investigation comprehensively explored the preparation, characterization, structure, stability, solubility, and bioavailability of the genistein-piperazine cocrystal. Cocrystal engineering provides a simple, efficient, and environmentally friendly approach to optimize the pharmaceutical properties of natural products for effective drug formulation and delivery. The study successfully prepared a novel cocrystal of genistein and piperazine, clarifying its structure and validating its stability. In vitro dissolution and in vivo pharmacokinetics studies demonstrated increased solubility, dissolution rate, and bioavailability. Overall, cocrystallization has enormous potential for optimizing the pharmaceutical properties of natural products to facilitate successful drug formulation and delivery.
